A shortage of a protein called TDP-43 caused muscle wasting and stunted nerve cells. This finding supports the idea that malfunction of this protein plays a decisive role in ALS and FTD. The study is published in the "Proceedings of the National Academy of Sciences of the USA" (PNAS).

ALS is an incurable neurological disease which manifests as rapidly progressing muscle wasting. Both limbs and respiratory muscles are affected. This leads to impaired mobility and breathing problems. Patients commonly die within a few years after the symptoms emerged. In rare cases, of which the British physicist Stephen Hawking is the most notable, patients can live with the disease for a long time. In Germany estimates show over 150,000 patients suffering from ALS an average of 1 in 500 people.

Proteins gone astray

Over the last few years, there has been increasing evidence that ALS and FTD a form of dementia associated with changes in personality and social behaviour may have similar or even the same origins. The symptoms overlap and common factors have also been found at the microscopic level. In many cases, particles accumulate and form clumps in the patient's nerve cells: this applies particularly to the TDP-43 protein.

"Normally, this protein is located in the cell nucleus and is involved in processing genetic information," explains molecular biologist Dr. Bettina Schmid, who works at the DZNE Munich site and at LMU. "However, in cases of disease, TDP-43 accumulates outside the nucleus forming aggregates." Schmid explains that it is not yet clear whether these clumps are harmful. "However, the protein's normal function is clearly disrupted. It no longer reaches the nucleus to perform its actual task. There seems to be a relationship between this malfunction and the disease."

Studies on zebrafish

However, until now little was known about the function of TDP-43. What are the consequences when this protein becomes non-functional? In order to answer this question, the team led by Bettina Schmid cooperated with the research group of Prof. Christian Haass to investigate the larvae of specially bred zebrafish. Their genetic code had been modified in such a way that no TDP-43 was produced in the organism of the fish. The result: the young fish showed massive muscle wasting and died a few days after hatching. Moreover, the extensions of the nerve cells which control the muscles were abnormal.

"To some extent, these are symptoms typical of ALS and FTD. Therefore, a loss of function of TDP-43 does seem to play a critical role in the disease," says Haass, Site Speaker of the DZNE Munich Site and chair of Metabolic Biochemistry at LMU.

The study revealed one more finding which surprised the researchers: the blood flow of the fish was massively disturbed. "It is well known that circulatory disorders play a part in other forms of dementia, notably in the case of Alzheimer's," says Haass. "We now want to investigate whether such problems with blood flow may be a general problem of neurodegenerative diseases and whether such problems occur particularly in patients with ALS and FTD."

Pentagon says it will resume F-35 flights

WASHINGTON (Reuters) - The Pentagon said on Thursday it would resume flights of its F-35 fighter jets following a week-long precautionary grounding imposed after a crack was found on an engine blade on a test plane in California.

"F-35 flight operations have been cleared to resume," Pentagon spokeswoman Kyra Hawn said in a statement. She said flights could begin as early as Friday, depending on weather conditions.

No additional cracks were found during inspections of engines on the remaining 50 planes in the Pentagon's fleet, or any spare engines, Hawn said.

The order allowing flights to resume is good news for the $396 billion F-35 Joint Strike Fighter, the Pentagon's biggest weapons program which is bracing for big cutbacks when automatic across-the-board budget cuts take effect on Friday.

It comes two days after the Pentagon's F-35 program chief blasted enginemaker Pratt & Whitney and prime contractor Lockheed Martin Corp for trying to "squeeze every nickel" out of the Pentagon and not shouldering enough risk on the program.

The order ends the program's second engine-related flight ban in less than two months. The Marines Corps version of the plane was grounded for nearly a month from mid-January because of a faulty hose in the engine, later blamed on manufacturing errors.

Matthew Bates, a spokesman for Pratt & Whitney, which supplies the engines for the planes, said the Pentagon's F-35 program office had decided to rescind the grounding order after extensive tests and analysis of the cracked turbine blade, which was discovered on February 19 during a routine inspection conducted after every 50 hours of flight time.

Bates said Pratt engineers had been working around the clock with Pentagon experts to determine the cause of the crack in the engine blade, including a "destructive" test that cracked open the blade.

The tests showed that the crack resulted from the "unique operating environment" in flight tests -- many of which tested the engine's powerful after burners -- rather than a high-cycle fatigue crack, which would have required a design change.

Bates said the engine in question had operated at high temperatures for more than four times longer than a typical F-35 flight, which led to a separation of the "grain boundary" of this particular blade.

The Navy order rescinding the flight ban, or so-called "red stripe," said that engine had experienced the most "hot engine time exposure" of all the engines in the developmental program. It said it would now require reports to monitor and limit similar damage after every 25 flight hours.

Pratt recommended the resumption of flights and ground operations of the engine earlier on Thursday after its engineers ruled out a high-cycle fatigue crack.

The Pentagon said the investigation concluded that the 0.6-inch long crack was caused by "prolonged exposure to high levels of heat and other operational stressors."

Pratt, a unit of United Technologies Corp, supplies the engine for the single-engine, single-seat fighter plane, which is built by Lockheed Martin Corp.

Contaminated diet contributes to exposure to endocrine-disrupting chemicals: Phthalates and BPA

Feb. 27, 2013 ? While water bottles may tout BPA-free labels and personal care products declare phthalates not among their ingredients, these assurances may not be enough.

According to a study published February 27 in the Nature Journal of Exposure Science and Environmental Epidemiology, we may be exposed to these chemicals in our diet, even if our diet is organic and we prepare, cook, and store foods in non-plastic containers. Children may be most vulnerable.

"Current information we give families may not be enough to reduce exposures," said Dr. Sheela Sathyanarayana, lead author on the study and an environmental health pediatrician in the UW School of Public Health and at Seattle Children's Research Institute. She is a physician at Harborview Medical Center's Pediatric Environmental Health Specialty Unit, and a UW assistant professor of pediatrics.

Phthalates and bisphenol A, better known as BPA, are synthetic endocrine-disrupting chemicals. Previous studies have linked prenatal exposure to phthalates to abnormalities in the male reproductive system. Associations have also been shown between fetal exposure to BPA and hyperactivity, anxiety, and depression in girls.

The researchers compared the chemical exposures of 10 families, half of whom were given written instructions on how to reduce phthalate and BPA exposures. They received handouts prepared by the national Pediatric Environmental Health Specialty Units, a network of experts on environmentally related health effects in children. The other families received a five-day catered diet of local, fresh, organic food that was not prepared, cooked or stored in plastic containers.

When the researchers tested the participants' urinary concentrations of metabolites for phthalates and BPA, they got surprising results. The researchers expected the levels of the metabolities to decrease in those adults and children eating the catered diet.

Instead, the opposite happened. The urinary concentration for phthalates were 100-fold higher than the those levels found in the majority of the general population. The comparison comes from a study conducted by the National Health and Nutrition Examination Survey. This is a program of studies managed by the Centers for Disease Control and Prevention and designed to assess the health and nutritional status of adults and children in the United States.

The concentrations were also much higher for children as compared to the adults. The researchers then tested the phthalate concentrations in the food ingredients used in the dietary intervention. Dairy products -- butter, cream, milk, and cheese -- had concentrations above 440 nanograms/gram. Ground cinnamon and cayenne pepper had concentrations above 700 ng/g, and ground coriander had concentrations of 21,400 ng/g.

"We were extremely surprised to see these results. We expected the concentrations to decrease significantly for the kids and parents in the catered diet group. Chemical contamination of foods can lead to concentrations higher than deemed safe by the US EPA," said Dr. Sheela Sathyanarayana.

Using the study results, the researchers estimated that the average child aged three to six years old was exposed to 183 milligrams per kilogram of their body weight per day. The U.S. Environmental Protection Agency's recommended limit is 20 mg/kg/day.

"It's difficult to control your exposure to these chemicals, even when you try," said Sathyanarayana. "We have very little control over what's in our food, including contaminants. Families can focus on buying fresh fruits and vegetables, foods that are not canned and are low in fat, but it may take new federal regulations to reduce exposures to these chemicals."

The other researchers in the study included Garry Alcedo (Seattle Children's Research Institute), Brian E. Saelens and Chuan Zhou (UW Department of Pediatrics, Seattle Children's Research Institute), Russell L. Dills and Jianbo Yu (UW Department of Environmental and Occupational Health Sciences) and Bruce Lanphear (BC Children's Hospital and Simon Fraser University).

Their paper is titled, "Unexpected results in a randomized dietary trial to reduce phthalate and bisphenol A exposure."

The study was supported through by the Center for Ecogenetics and Environmental Health in the Department of Environmental and Occupational Health Sciences in the UW School of Public Health. A grant from the National Institute of Environmental Health provides major support for the center.

At the RSA Conference today, I attended an excellent panel discussion titled Y U NO HAZ METRICS? The speakers were David Mortman, Jack Jones, Alex Hutton, and Caroline Wong, and the panel was was moderated by John Johnson. The panel discussed risk management more than they discussed specific metrics, which was slightly different than what I expected. However, the panel surpassed my expectations.

A commenter towards the end of the session made an analogy which I thought was a good one. He said that risk management is like risotto. It has three basic ingredients, and you put them together and adjust the balance until it tastes good to you. In other words, no two risottos (or risk management programs) will ever be exactly the same. The commenter then brought up this follow-up concern: that we as an industry don?t even have an agreed-upon list of standard ingredients for our risk management risotto.

There are different risk management frameworks, which use and rely on different ingredients. However I think that Alex Hutton said it well:

Metrics are a critical part of a risk management system. Without metrics, it is impossible to tell whether the mitigation techniques and controls are working. In today?s world, you have an infinite number of threats and concerns to be worried about, and a fixed amount of time, money, and energy that can be spent. Without metrics, how can you determine where to spend that time, money, and energy? And how can you justify that the time, money, and energy was spent in the right place when you are served with a lawsuit because of a breach?

The last and possibly most important takeaway I?ll provide is this: useful metrics must relate to business value. Typically, risk management systems include metrics such as percentage of systems that are fully patched or amount of time it takes to apply a patch. These on their own are not useful metrics to a risk management system. They are only useful if they are tied back to things that are important to the business. So consider instead that the business impact of unpatched systems is that you could have a virus outbreak, and spend days restoring systems and handling the PR nightmare you?ve created. Or consider the business impact of having your e-commerce site down for three hours while you deploy patches.
